1. Technical means of SLOAD

We use Java to write the underlying code, HTML5 for front-end page design; SLOAD is based on the MySQL database running on the cloud server.


2. The results provided by SLOAD

More than 100,000 gene pairs with synthetic lethal effects were predicted in 31 cancer types, achieving a mean AUC of 0.816. This high quality combination of genes with synthetic lethal effects can provide more reliable drug targets.


3. Methods of data analysis

3.1 Genetic mutations

Based on the potential role of gene mutation in the synthetic lethal effect, we first analyzed the gene mutation rate in 33 cancers.The mutation rate of pairs of genes is the percentage of the samples where the combination of genes was mutated in each cancer.The results show that there are differences in mutation rates of genes in different cancers, and there are also differences in mutation rates of different genes in the same cancer.

3.2 Differential Analysis

LogFC and Padj were calculated using EdgeR packages based on mRNA expression data from different cancer types (only data containing controls were analyzed). The number of paracancer samples in TCGA database is small, and some cancer types lack paracancer samples. In order to obtain more accurate characteristic values, the combined TCGA and GTEX data provided by XENA database are used for differential expression analysis.Some results are shown behind.


4. Example Usage

The users can comprehensively browse cancer-specific synthetic lethal interactions and corresponding multiple-omics results.
